The ribosomal ribonucleic acids (rRNAs) of Spiroplasma citri and corn stunt spiroplasma (CSS) were compared with rRNAs of Acholeplasma laidlawii, Mycoplasma hominis, Bacillus subtilis, and Escherichia coli by polyacrylamide gel electrophoresis. In nondenaturing gels, the 23s rRNA of spiroplasmas and B. subtilis migrated faster than the 23s rRNA of A. laidlawii and E. coli, but the 16s rRNAs comigrated. In denaturing gels, the 16s rRNAs behaved similarly but the 23s rRNAs of spiroplasmas and A. laidlawii migrated faster than their bacterial counterpart. The 5s rRNA of S. citri and CSS was 35000 daltons, intermediate in size between 5s rRNAs of A. laidlawii (36500 daltons) and M. hominis (32200 daltons). The 5s rRNA of A. laidlawii was, in turn, smaller than the bacterial 5s rRNAs. It is concluded that mycoplasmas have the smallest 5s rRNA reported.; Deoxyribonucleic acid hybridization studies were performed to assess the degree of genetic relatedness between CSS and S. citri. At 56(DEGREES)C, the degree of homology between CSS and S. citri was about 68% (63 to 75%). The level of homology among four S. citri isolates tested ranged from 93 to 100%. The protein patterns of S. citri isolates were identical on polyacrylamide gels but partial similarity was observed between CSS and S. citri. It is suggested that CSS and S. citri belong to the same species but to separate subspecies or varieties.
