摘要

[ ABSTRACT] AIM:To investigate the effect of DEK downregulation on the apoptosis of gastric carcinoma SGC-7901 cells, and to explore its associations with NF-κB signaling pathway and apoptosis related proteins.METHODS:SGC-7901 cells with different treatments were divided into 3 groups including untreated group, control siRNA group and DEK siRNA group.The expression of DEK at mRNA and protein levels in the SGC-7901 cells was detected by real-time PCR and Western blot.The cell apoptosis was examined by flow cytometry.Furthermore, the activities of caspase-3 and caspase-9 in the SGC-7901 cells were investigated by Caspase-Glo?-3/9 kit.Finally, the expression of key regulatory pro-tein p65 of NF-κB signaling pathway and apoptosis-related proteins Bcl-2 and Bax in the SGC-7901 cells was investigated by Western blot.RESULTS:Compared with untreated group and control siRNA group, the expression of DEK at mRNA and protein levels was significantly downregulated in DEK siRNA group (P<0.05).In addition, the ratios of early phase apoptosis and total apoptosis in DEK siRNA group were markedly higher than those in untreated group and control siRNA group (P<0.05).Most notably, the decrease in p65 and Bcl-2 proteins, increase in Bax protein and the increases of caspase-3 and caspase-9 activities were observed in DEK siRNA group.CONCLUSION:Downregulation of DEK mediates cell apoptosis of gastric carcinoma may be tightly associated with NF-κB signaling pathway.%目的:研究DEK表達(dá)下調(diào)對(duì)胃癌SGC-7901細(xì)胞凋亡的影響,并分析其與NF-κB信號(hào)途徑及凋亡相關(guān)蛋白表達(dá)的關(guān)系。方法:將DEK siRNA和對(duì)照siRNA轉(zhuǎn)染胃癌SGC-7901細(xì)胞,并將SGC-7901細(xì)胞分為未處理組、對(duì)照siRNA組及DEK siRNA組。采用實(shí)時(shí)熒光定量PCR和Western blot檢測(cè)3組胃癌SGC-7901細(xì)胞中DEK mRNA和蛋白的表達(dá);流式細(xì)胞術(shù)檢測(cè)3組SGC-7901細(xì)胞凋亡的變化;Caspase-Glo?-3/9試劑盒檢測(cè)3組SGC-7901細(xì)胞中caspase-3和caspase-9的活性;Western blot技術(shù)檢測(cè)3組SGC-7901細(xì)胞中NF-κB信號(hào)途徑關(guān)鍵調(diào)控蛋白p65及凋亡相關(guān)蛋白Bcl-2和Bax的表達(dá)。結(jié)果:與未處理組和對(duì)照siRNA組相比,DEK siRNA組SGC-7901細(xì)胞中DEK的mRNA和蛋白表達(dá)水平顯著下調(diào)(P<0.05)。 DEK siRNA組SGC-7901細(xì)胞的早期凋亡率和總凋亡率均顯著高于未處理組和對(duì)照siRNA組( P<0.05)。最為顯著的是,DEK siRNA轉(zhuǎn)染細(xì)胞后,p65和Bcl-2蛋白表達(dá)下調(diào),Bax蛋白表達(dá)上調(diào),并伴隨caspase-3和caspase-9活性上升。結(jié)論:DEK表達(dá)下調(diào)介導(dǎo)的SGC-7901細(xì)胞凋亡可能與NF-κB信號(hào)途徑密切相關(guān)。
