    • 摘要: 從鎘污染礦區(qū)水體污泥中分離篩選得到一株對19 mmol/L鎘(Cd)有抗性的菌株L202002,通過形態(tài)觀察、生化和生理特性及ITS序列分析,對該菌株進(jìn)行了鑒定,考察了該菌株對高嶺土懸濁液的絮凝活性和對重金屬Cd、Cu、Cr、Hg、Pb、Zn的抗性及去除溶液中Cd的能力.結(jié)果表明:該菌株屬于曲霉菌(A spergillus),與土曲霉(Aspergillus cejpii)的同源性大于98%;對多種重金屬有較強(qiáng)的抗性,在固體培養(yǎng)基中的抗性順序為Cd>Zn>Cu>Cr>Hg>Pb;菌株對高嶺土懸濁液有很好的絮凝活性;活細(xì)胞和非活細(xì)胞都能去除溶液中的Cd,且活細(xì)胞比非活細(xì)胞具有更好的生物吸附能力.該菌株在含重金屬廢水處理中有應(yīng)用潛能.

    • 摘要: 采用常規(guī)的平板劃線分離法,搖床發(fā)酵活性污泥及附近土壤中篩選高效產(chǎn)絮凝劑菌株,結(jié)合形態(tài)學(xué)觀察及分子生物學(xué)對其進(jìn)行鑒定.以豆制品廢水為培養(yǎng)基,優(yōu)化菌株發(fā)酵產(chǎn)絮凝劑培養(yǎng)條件,并對絮凝活性成分進(jìn)行分析表征.結(jié)果表明,從豆制品廢水活性污泥及附近土壤樣品中分離得到一株高效的產(chǎn)絮菌,編號為J-7,經(jīng)形態(tài)學(xué)及16S rDNA基因測序,菌株J-7被鑒定為Talaromyces pinophilus.最佳培養(yǎng)條件為廢水添加量為60%、接種量4%、培養(yǎng)溫度為25°C、廢水初始pH值為5、搖床轉(zhuǎn)速為150 r/min、發(fā)酵時間48 h.在該優(yōu)化條件下,絮凝率可達(dá)93.42%.絮凝活性成分進(jìn)行定性測定、紫外光譜掃描以及紅外光譜分析,初步判斷其主要成分為多糖,主要基團(tuán)包括羰基、羥基、氨基、碳骨架基團(tuán)等.

    • 摘要: 目的:獲得高效廉價的微生物絮凝劑.方法:利用常規(guī)篩選技術(shù)從宜州龍江河沿岸排污口污泥中分離篩選菌株,菌種鑒定是經(jīng)形態(tài)觀察、理化特性分析以及分子水平16S rDNA系統(tǒng)進(jìn)化分析進(jìn)行的;按照常規(guī)方法考察了絮凝劑在高嶺土、活性炭及酵母懸浮液中的絮凝性能和影響絮凝性能的因素.結(jié)果:獲得一株產(chǎn)絮凝劑的菌株L2019-2,該絮凝活性菌株鑒定為維羅納假單胞菌(Pseudomonas veronii).其產(chǎn)生的生物高分子絮凝劑在無機(jī)懸浮液和有機(jī)懸浮液中均具有絮凝活性.在高嶺土懸浮液中,pH為5.0、溫度為30°C、助凝劑為Fe3+時,絮凝劑的絮凝活性較高,絮凝率迭98.30%.經(jīng)化學(xué)組成分析,該絮凝劑為酸性糖蛋白.結(jié)論:L2019-2所產(chǎn)絮凝劑具有很好的絮凝性能,可為研究微生物絮凝劑提供良好的菌種資源,在污水處理中更具有很好的應(yīng)用潛力.

    • 摘要: 為將具有絮凝淀粉活性的酸漿應(yīng)用在玉米淀粉生產(chǎn)中,試驗采用從自然發(fā)酵甘薯酸漿中分離篩選得到1株對淀粉顆粒具有絮凝活性的副干酪乳桿菌副干酪亞種L1(Lactobacillus paracasei subsp.paracasei L1)接種在玉米漿中發(fā)酵制成玉米酸漿,研究不同氮源與碳源、不同接種量、培養(yǎng)時間、培養(yǎng)溫度、培養(yǎng)pH值等條件對玉米酸漿絮凝淀粉活性的影響.試驗采用不同發(fā)酵條件制備玉米酸漿,以5%的比例將玉米酸漿加入到玉米淀粉漿(5%,w/v)中,靜置3min,采用碘顯色法測定上清液液面下20mL處的淀粉含量,上清液中玉米淀粉含量越低則表明玉米酸漿的絮凝活性越好.結(jié)果表明:當(dāng)酵母浸粉為氮源,乳糖為碳源時,玉米酸漿沉降淀粉的能力較強(qiáng),酵母浸粉的最佳用量為3.0%,乳糖為2%,繼續(xù)增加酵母浸粉及乳糖的添加量,絮凝活性反而降低;當(dāng)接種量為10%時,玉米酸漿的絮凝活性最好.玉米酸漿培養(yǎng)36h時,玉米酸漿的凝絮活性較高,為0.10mg?mL-1;隨著培養(yǎng)溫度的升高,玉米酸漿的絮凝活性先升高后降低,30°C時玉米酸漿的絮凝活性最大;酸漿絮凝活性在pH值5.5~6.0時較好,隨著pH值的升高,絮凝活性降低;玉米粒浸泡時間24h時,玉米酸漿絮凝淀粉的能力較強(qiáng);玉米以1:3的比例加水磨漿時,玉米酸漿絮凝活性最好.由此可以看出,不同發(fā)酵條件對玉米酸漿絮凝淀粉活性的影響較大,玉米酸漿的最佳培養(yǎng)方法為玉米粒浸泡24h,以1:3的比例加水磨漿過濾后,加入3.0%酵母浸粉、2%乳糖,接種10%的副干酪乳桿菌副干酪亞種L1發(fā)酵液,調(diào)整pH值5.5~6.0,30°C培養(yǎng)36h.

    • 摘要: [目的]研究膠質(zhì)芽孢桿菌(Bacillus mucilaginous)MY6-2的絮凝特性及其對重金屬Pb2+的吸附作用,為微生物絮凝劑的實際應(yīng)用提供參考.[方法]通過發(fā)酵、醇沉、透析、冷凍干燥得到膠質(zhì)芽孢桿菌MY6-2生物絮凝劑;研究pH、絮凝劑用量、金屬助凝劑及其用量、加熱對MY6-2絮凝劑活性的影響;分析溶液pH、Pb2+初始質(zhì)量濃度及絮凝劑用量對MY6-2絮凝劑吸附Pb2+的影響.[結(jié)果]MY6-2絮凝劑的主要成分為多糖,占樣品的65.68%;最佳絮凝條件為:pH>7.0,絮凝劑用量20 mL/L,助凝劑金屬離子為Ca2+(質(zhì)量分?jǐn)?shù)1%,用量為20 mL/L),在此條件下MY6-2絮凝活性可達(dá)到95%以上.在100°C水浴中加熱30 min絮凝活性無明顯變化,表明該絮凝劑具有良好的熱穩(wěn)定性.MY6-2絮凝劑對Pb2+的最佳吸附條件為:pH4.0、Pb2+初始質(zhì)量濃度100~500 mg/L、絮凝劑用量80mL/L,在此條件下,MY6-2絮凝劑對Pb2+的絮凝活性均在80%以上.[結(jié)論]MY6-2絮凝劑具有較高的絮凝活性,對重金屬Pb2+具有較高的吸附活性,且生產(chǎn)成本低,安全性高,具有較高的推廣應(yīng)用價值.%[Objective] This study explored the characteristics of a microbial flocculant produced by Bacillus mucilaginosus MY6-2 and its adsorption of Pb2+ to provide basis for actual applications.[Method] A novel bio-flocculant was obtained from Bacillus mucilaginosus MY6-2 by alcohol precipitation,dialysis and lyophilization.The effects of pH,MBF dosage,cation supplement and temperature on flocculating activity were studied.The adsorption of Pb2+ was tested using batch method with respect to pH,initial metal ion concentration and MBF dosage.[Result] The main component of MBF MY6-2 was extracellular polysaccharide.Under the optimum condition of pH>7.0,MBF dosage 20 mL/L Kaolin suspension,and 1% CaCl2 20 mL/L,the obtained flocculating activity was >95 %.The flocculating activity remained after being heated to 100 °C for 30 min,indicating that MBF MY6-2 had high thermal stability.The optimum adsorption conditions were pH 4.0,initial ion concentration 100-500 mg/L and MBF dosage 80 mL/L.The adsorption rate was maintained above 80% under these conditions.[Conclusion] MBF MY6-2 is a cost-effective and safe microbial flocculant,and it has high flocculating activity and adsorption activity of Pb2+.

    • 摘要: By natural fermentation mung bean sour liquid as the research object,using plate screening techniques isolated five strains with starch flocculation activity of acid producing bacteria from sour liquid.By group morphology,individual morphological observation,physiological and biochemical experiments and 16S rDNA sequence analysis strain LLY11 with the highest flocculation activity were identificated.The strain was identified as Acetobacter indonesiensis and named as Acetobacter indonesiensis LLY11.The culture conditions were optimized by single factor experiments,the best culture condition was glucose 1%,beef paste 2%,initial pH5.0,culture temperature 30 °C,under this culture condition,the growth curve of LLY11 strain was determined.h was determined that the strain began to enter logarithmic period from 12 h,appeard secondary growth and reached the highest value at 24 h and 42 h,and then entered the decline stage.%以自然發(fā)酵綠豆酸漿為研究對象,采用平板分離技術(shù)從酸漿中分離出5株具有絮凝淀粉活性的產(chǎn)酸細(xì)菌,通過群體形態(tài)、個體形態(tài)觀察、生理生化實驗及16S rDNA序列分析對絮凝活性最高的菌株LLY11進(jìn)行菌種鑒定,鑒定該菌株為Acetobacter indonesiensis,并命名為Acetobacter indonesiensis LLY11.通過單因素實驗優(yōu)化了其培養(yǎng)條件,其最佳培養(yǎng)條件為:分別以1%葡萄糖、2%牛肉膏作為綠豆汁培養(yǎng)基的碳源和氮源、初始pH5.0、培養(yǎng)溫度30°C,在該培養(yǎng)條件下,測定了LLY11菌株的生長曲線,確定菌株從12h才開始進(jìn)入對數(shù)期,有二次生長,分別在24 h和42 h菌體數(shù)量達(dá)到最高值,隨后進(jìn)入衰亡期.

    • 摘要: Using marine Bacillus licheniformis DHS-40 as a flocculant-producing strain,we determined the active components of microbial flocculant by a qualitative experiment,evaluated flocculation stability by a sixcombined agitator for program-controlled coagulation test and a gravity sedimentation method,characterized microscopic morphologies of microbial flocculant and flocculant particles by scanning electron microscope and atomic force microscope,and discussed flocculation mechanism.Results indicated that the active components of microbial flocculant of marine Bacillus licheniformis were mixture of polysaccharides and proteins.The flocculation activity was above 80% when microbial flocculant dosage was 0.05 g · L-1,environmental pH value was 4.0~8.0,and environmental temperature was 0~50 °C.Environmental pH value hardly affect the flocculation activity,however,the flocculation activity decreased rapidly under the alkaline environment(pH>8.0),and lost all when pH value was 10.0.The morphology of microbial flocculant possessed a chain structure with different depths and channels.The morphology of flocculant particles presented a stacked structure.Depended on charge adsorption and bridging action,microbial flocculant was settled together with the turbidity of water,thus a chieved the goal of purifying water body.%以海洋地衣芽孢桿菌DHS-40為絮凝劑產(chǎn)生菌,通過定性測試鑒定所產(chǎn)微生物絮凝劑的絮凝活性有效成分,采用程控混凝實驗六聯(lián)攪拌儀和重力沉降法評價其絮凝穩(wěn)定性,通過掃描電子顯微鏡和原子力顯微鏡表征微生物絮凝劑和絮凝顆粒的微觀形貌,并初步探討了微生物絮凝劑的絮凝機(jī)理.結(jié)果表明,海洋地衣芽孢桿菌DHS-40所產(chǎn)微生物絮凝劑的絮凝活性有效成分為多糖和蛋白質(zhì)的混合物;當(dāng)微生物絮凝劑投加量為0.05 9·L-1、環(huán)境pH值為4.0~8.0、環(huán)境溫度為0~50°C時,絮凝活性達(dá)80%以上;絮凝活性基本不受環(huán)境pH值的影響,但在堿性較強(qiáng)(pH>8.0)時絮凝活性迅速下降,在pH值為10.0時完全喪失;微生物絮凝劑呈鏈狀結(jié)構(gòu),且有深淺不等的溝回;絮凝顆粒呈層疊結(jié)構(gòu).微生物絮凝劑依靠電荷吸附和橋聯(lián)作用帶動水中的渾濁物共同沉降,從而達(dá)到凈化水體的目的.

    • 摘要: 為了篩選針對畜禽養(yǎng)殖污水的高效絮凝菌,試驗通過富集、分離、純化和絮凝活性測定,從紹興縣中大畜牧有限公司(生豬飼養(yǎng)繁育)氧化塘的污水和淺層底泥中篩選絮凝菌,通過形態(tài)學(xué)、生理生化反應(yīng)和16S rDNA測序,對絮凝能力最強(qiáng)的菌株進(jìn)行鑒定.結(jié)果表明:從養(yǎng)豬場氧化塘的污水和淺層底泥中分離篩選到7株絮凝菌,其中C4絮凝活性最強(qiáng),鑒定為解木糖賴氨酸芽胞桿菌(Lysiniba-cillus xylanilyticus).

    • 摘要: An efficiently flocculant-producing bacterial strain WX-7 has been screened out and obtained as Micro-bacterium sp.,through 16s rDNA determination. It grows best under the following conditions:pH is 8.0,inoculum concentration 1%,temperature 30°C,and rotational speed 170 r/min,by adding sucrose and dried corn steep liquor powder and culturing for 16-24 h. The flocculating activity of Strain WX-7 mainly distributes on thallus. The best coagulant aid is Ca2+. When pH is higher than 11,and 3 mL of bacterial liquid,and 3 mL of CaCl2 solution (1%) are added to kaolin suspension(0.5%),the maximum flocculating activity can reach 98%. The flocculant ingredient is not sensitive about heat treatment. The flocculant produced from Strain WX-7 can effectively remove the chroma and COD from dyeing wastewater. The decolorization rate and COD removing rate are 81%and 42%,respectively. It indicates that this strain has certain application prospect in wastewater treatment fields.%篩選獲得一株高效產(chǎn)絮凝劑菌株WX-7,通過16s rDNA鑒定,該菌株隸屬于微桿菌屬。在pH為8.0、接種量1%、30°C、170 r/min條件下,添加蔗糖、玉米漿干粉,培養(yǎng)16~24 h生長最佳。 WX-7菌株的絮凝活性主要分布于菌體,最佳助凝劑為Ca2+。在pH>11條件下,向0.5%的高嶺土懸液投加3 mL菌液和3 mL 1%的CaCl2溶液,絮凝活性最高可達(dá)98%。絮凝劑成分對熱處理不敏感。 WX-7菌株所產(chǎn)絮凝劑能有效去除印染廢水的色度和COD,脫色率和COD去除率分別為81%、42%,在廢水處理中具有一定的應(yīng)用前景。

    • 摘要: 利用啤酒廢水培養(yǎng)微生物絮凝劑產(chǎn)生菌Bacillus simplex PS1,通過單因素試驗和正交試驗,優(yōu)化其利用啤酒廢水產(chǎn)生絮凝劑的培養(yǎng)條件;并考察所產(chǎn)絮凝劑對實際廢水的絮凝效果.結(jié)果表明,Bacillus simplex PS1以啤酒廢水為原料產(chǎn)生微生物絮凝劑的最佳培養(yǎng)條件為:啤酒廢水COD濃度5 000 mg/L、KH2P041 g/L、K2HPO42.5 g/L、初始pH值7、接種量2%(體積比)、培養(yǎng)溫度30°C、搖床轉(zhuǎn)速170 r/min、培養(yǎng)時間36 h.在此培養(yǎng)條件下,菌株所產(chǎn)絮凝劑對高嶺土懸濁液的絮凝率達(dá)到96.8%,對城市污水、乳品廢水、醫(yī)院廢水、淀粉廢水、餐飲廢水均具有良好的凈化效果,對廢水濁度去除率達(dá)到90%以上,對COD和色度的去除率達(dá)到80%以上,表明Bacillus simplex PS1利用啤酒廢水所產(chǎn)微生物絮凝劑處理廢水是完全可行的.
